چکیده

Piperine is a dietary alkaloid with huge pharmacological importance and was first isolated from pepper extract. It has been reported anti-cancer, anti-amoebic, anti-mycobacterial, anti-mutagenic, anticonvulsant, anti-asthmatic, immunomodulatory, anti-inflammatory anti-oxidant activity. Therefore, the current investigation designed to unveil anticancer effects of piperine molecule against human breast carcinoma. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay executed for determination on cancer cell viability. Clonogenic implemented checking effect clonogenic potency cells. To monitor morphology cancerous cells, phase contrast microscopy performed after exposure. Apoptosis checked by acridine orange/ethidium bromine staining execution Western blotting analysis. Cell migration invasion monitored transwell chambers assay. Muse flow cytometric analysis assessing different cycle phases. check levels phosphatidylinositol 3-kinase protein kinase B. Results revealed that induced dose reliant cytotoxicity in MCF-7 Further, application colonies remarkably reduced number minimum. Phase evidenced some specific morphological modifications indicating apoptosis allied like apoptotic bodies, condensed nucleus membrane blabbing. Acridine cells which further investigated blotting. This increased expression B-cell lymphoma 2 associated X 2, induction piperine. as well invasive ability minimum dependent-manner. Flow arrested at growth 2/mitosis phase, hence suppressed progression. Finally, predicted constant B phosphorylated 3-kinase. Hence, blocking 3-kinase/protein signaling pathway. In conclusion, regarding remarkable suppression apoptosis, invasion, blocked 3-kinase/

Targeting the phosphatidylinositol 3-kinase signaling pathway in breast cancer.

The phosphatidylinositol 3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR) network plays a key regulatory function in cell survival, proliferation, migration, metabolism, angiogenesis, and apoptosis. Genetic aberrations found at different levels, either with activation of oncogenes or inactivation of tumor suppressors, make this pathway one of the most commonly disrupted in human breast ...

Targeting the phosphatidylinositol 3-kinase/Akt pathway for enhancing breast cancer cells to radiotherapy.

The phosphatidylinositol 3-kinase (PI-3K)/Akt pathway, regulated by its upstream growth factor receptor tyrosine kinases, plays a critical role in promoting cell proliferation and inhibiting cell death. The aim of this study was to determine whether the PI-3K/Akt activity contributes to the resistance of human breast cancer cells to ionizing radiation and whether inhibition of the PI-3K/Akt pat...

β-inducible gene-h3 promotes human breast carcinoma cell metastasis by activating the phosphatidylinositol 3-kinase/protein kinase B signaling pathway

Metastatic breast cancer is one of the most common metastatic tumors. Although studies have validated the role of β-inducible gene-h3 (βig-h3) in human biology and disease, the detailed mechanisms mediated by βig-h3 in breast carcinoma metastasis remain unclear. Thus, the present study investigated the role and potential mechanism of βig-h3 during breast carcinoma cell metastasis. The results i...
